El juego se centra en distintos personajes de franquicias de juegos de Nintendo, tales como Mario Bros, Kirby, Pokémon y The Legend of Zelda. Los escenarios y modos de juego hacen referencia, o toman sus diseños desde juegos populares lanzados por Nintendo. El sistema de juego de Melee ofrece un acercamiento poco ortodoxo al género de los juegos de lucha, puesto que el daño recibido se mide con contadores de porcentaje, en vez de las tradicionales barras de energía vistas en la mayoría de los juegos del género. Melee amplía el encanto de su predecesor al incluir nuevos modos de juego y personajes controlables.
